Research and Applications of Interleukins

Interleukins (ILs) are cytokines produced by white blood cells or other cell types, which transmit signals between cells, regulating cell growth, differentiation, and immune function. Initially identified as being primarily produced by and acting on white blood cells, they were named ‘interleukins’ (ILs).
ILs play a pivotal role in the immune system, exhibiting broad biological functions and great significance in medical research and clinical applications. Below are their primary roles and research applications:
1. Main Biological Functions of Interleukins
1.1 Immune Regulation
Regulate the proliferation, differentiation, and activation of immune cells. For example, interleukin-2 (IL-2) promotes the proliferation and differentiation of T cells, enhancing cellular immune function.
Regulate the interactions between immune cells and mediate their migration and homing. For example, IL-8 recruits immune cells such as neutrophils to migrate to sites of inflammation.
1.2 Inflammatory Response
Act as inflammatory mediators to participate in the initiation and maintenance of inflammation. For example, IL-1, IL-6, and other interleukins stimulate the production of acute phase reaction in the body, causing symptoms such as fever, warmth, redness, swelling, and pain at inflammatory sites.
Regulate the intensity and duration of inflammatory responses to prevent damage to the body from excessive inflammation. For instance, IL-10 exerts anti-inflammatory effects by inhibiting the production of various pro-inflammatory cytokines.
1.3 Regulation of Hematopoietic Function
Regulate the proliferation and differentiation of hematopoietic stem cells (HSCs) and promote the production of various blood cell types. For example, IL-3 acts on pluripotent hematopoietic stem cells, promoting their differentiation into multiple blood cell lineages.
 
2. Research Applications of Interleukins
2.1 Disease Diagnosis and Monitoring
Changes in the levels of certain interleukins can serve as auxiliary indicators for disease diagnosis. For example, in infectious diseases, serum levels of IL-6, IL-10, and other interleukins are often significantly elevated, helping assess the severity of infection and prognosis.
Monitoring the dynamic changes of interleukins during disease treatment aids in evaluating therapeutic efficacy and predicting the risk of disease recurrence. For instance, in tumor treatment, tracking the levels of relevant interleukins in patients' serum provides insights into changes in immune function and tumor progression.
2.2 Drug R&D
Target interleukins (ILs) and their receptors for drug development. For example, monoclonal antibodies (mAbs) against the IL-6 receptor have been used to treat autoimmune diseases such as rheumatoid arthritis (RA). They inhibit inflammatory responses by blocking the binding of IL-6 to its receptor.
Leverage the immune regulatory effects of ILs to develop immunotherapeutic drugs. For instance, recombinant IL-2 has been applied in tumor immunotherapy, activating the body’s anti-tumor immune response.
2.3 Cell Therapy
In cell therapy, interleukins (ILs) can be used for the in vitro culture and expansion of immune cells. For example, in CAR-T cell therapy, cytokines such as IL-2 promote the proliferation and activation of CAR-T cells, enhancing their anti-tumor activity.
Studies have shown that certain genetically modified cells expressing interleukins can be used to treat specific diseases. Genes encoding specific ILs are introduced into tumor cells or immune cells, which are then infused back into patients' bodies to enhance the anti-tumor immune response or regulate immune function.
